A Novel Composite Nano-scaffold with Potential Usage as Skin Dermo-epidermal Grafts for Chronic Wound Treatment

Background: Nano-scaffolds loaded with bioactive compounds such as ZnO nanoparticles (ZnO-NPs), tissue-engineered artificial skin grafts, can be a suitable substitute for extracellular matrix and greatly contribute to accelerating chronic wound treatment by decreasing the chance of bacterial infection. Methods: Silk fibroin nanofiber was fabricated using electrospinning three-dimensional porous hybrids (3DPH) nano-scaffold composite sodium alginate/ZnO-NPs solution silk electrospun nanofibers adopting freeze-drying method. Successful configuration nano-scaffolds were confirmed field emission scanning electron microscopy (FE-SEM). Antimicrobial activity, cell attachment, cytotoxicity evaluation scaffolds performed employing disk diffusion method, L929 culture, MTT assay, respectively. Results: Antibacterial analysis 3DPH revealed their appropriate antibacterial activity against Staphylococcus aureus Escherichia coli bacteria. Furthermore, results from adhesion analyses indicated viability, proliferation on nano-scaffold, which are fundamental healing dermo-epidermal grafts. Conclusions: In sum, an epidermal graft ZnO-NPs dermal fabricated. Moreover, 1.5% (w/v) concentrations selected incorporated into nano-scaffold. Considering promising biological analyses, nanofibrous may have been grafts regeneration.
